Ethephon (Ethrel®) is an ethylene-based plant growth regulator that used in agriculture and it has direct and indirect effects on human health, direct effect via its inhalation during usage in agriculture and indirect effect through the diet (Fruits and vegetables) that is sprayed with it. The current study aimed to examine the possible modifying effects of costus (Saussurea lappa) root aqueous extract against Ethephon induced liver toxicity, injury, DNA fragmentation and PCNA alterations in male rats. Fifty adult male rats were divided into 5 groups (1st, control; 2nd, Costus; 3rd, Ethephon; 4th, Post treated Ethephon with costus; 5th, self-healing Ethephon). Current results revealed that; a significant increase in aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), liver injury, DNA damage and PCNA expressions in Ethephon group when compared with control group. In contrast; a significant decrease in albumin and total proteins in Ethephon group when compared with control group. Treatment of rats with costus after Ethephon improved these alterations as compared with Ethephon self-healing. So, it could be concluded that costus root extract worth to be considered as a natural substance for ameliorating the hepatic toxicity induced by plant growth regulator Ethephon.

Objectives: Ginkgo biloba leaf extract (GBLE) and L-carnitine extract are a commercial product used as a nutraceutic herbal widely. They have antioxidant and reactive oxygen species (ROS) scavenger. This study hypothesized that GB and L-carnitine may have protective effects against pentylenetetrazol (PTZ) induced oxidative damage in kidney in male albino rats. Materials and Methods: A total of 80 male albino rats were equally divided into eight groups (G1, control; G2, GBLE; G3, L-carnitine; G4, Pentylenetetrazole; G5&G6, pre and post treated PTZ with L-carnitine groups respectively; G7&G8, pre and post treated PTZ with GBLE respectively). Results: Serum urea, creatinine, uric acid and cystatin c were significantly increased in PTZ group when compared with control. Also; kidney homogenate MDA and DNA fragmentation were increased when compared with control while, significant decrease in CAT, SOD, GST and TAC in PTZ when compared with control. On other hand pre and post-treatment with L-carnitine and GBLE improved these parameters. Conclusions: The results revealed that; the treatment with L-carnitine improved these parameters more than GBLE and the results of post treatment were better than pre-treatment.

Objectives: Thyroid hormones regulate all metabolic activities; therefore, it is not amazing that hepatic; renal dysfunctions and lipid alterations are commonly detected in patients with thyroid disorders. This study estimated some biochemical changes in post pubertal hyperthyroid and hypothyroid mice and their impacts on liver and kidney functions and also on changes in the lipid pattern. In addition, the ameliorating role of Costus root extract supplementation was examined. Material and Methods: A total of 60 male Swiss albino mice were randomly divided into 5 groups; control, control with costus extract, hypo- and hyper-thyroids post-treated with costus root extract. Results: Present results revealed that, a significant increase in serum thyrotropin (TSH), alanine transaminase (ALT) and aspartate transaminase (AST), alkaline phosphatase (ALP), creatinine, urea, potassium, chloride, cholesterol and triglycerides levels in hypothyroid mice compared to control. On the other hand; a significant decrease in serumthyroxine (T4), tri‐iodothyronine (T3), albumin, total protein and calcium ions levels in hypothyroid mice compared to control. In contrast to hypothyroidism, a significant increase in serum T3, ALT, AST, ALP, creatinine, urea, sodium, potassium, chloride and total proteins levels in hyperthyroid mice compared to control. On the other hand; a significant decrease in serum TSH, T4, albumin, calcium ions cholesterol and triglycerides levels in hyperthyroid mice compared to control. Treatments of mice with Costus root extract in both hypo- and hyperthyroidism modulates the measured serum parameters. Conclusions: Our results could propose that the extract of Costus roots can be used as an adjuvant co-therapy in hypo- and hyperthyroidism syndromes with propylthiouracil and Eltroxin replacement therapy, respectively

ABSTRACT Equigan is an anabolic steroid that has been developed for veterinary use and derived from endogenous sex hormone testosterone that plays a key role in the development of male reproductive tissue as well as in puberty and spermatogenesis. The current study is aimed to investigate the possible prophylactic effect of star anise extracts (SAE) on the toxicity of rat testes, sexual hormones alternations, sperm count, sperm abnormalities and testicular DNA damage by Equigan. Forty adult male rats were equally divided into four groups (1st Control group, 2nd SAE group, 3rd Equigan and 4th Equigan+SAE group). Food and fluid intakes, relative body weight, potassium, chloride, phosphorous, non-progressive and immotile sperms were significantly increased in Equigan group as compared to control group. In contrast; relative testes weight, sodium, magnesium, total calcium, testosterone, FSH, LH, PRL, sperm count, progressive motility, and viability showed a significant decrease in Equigan group as compared to control groups. The relative weight of epididymis, seminal vesicles, prostates and serum calcium ions didn't change significantly in different studied groups. Co-administration of SAE with Equigan improved the sexual toxicity, electrolyte alternations, sperm count, abnormalities and DNA damage induced by Equigan.

ABSTRACT Gleevec (imatinib) is an antineoplastic chemotherapeutic agent used in the treatment of many types of cancer. The current study was conducted to examine the possible modifying effects of grape seeds proanthocyandins extract (GSPE) against apoptosis, liver injury and Ki67 alterations induced by Gleevec in male rats. 40 male albino rats were equally divided into four groups (First and second groups were control and GSPE groups; third group was Gleevec group and fourth group was treated with Gleevec and GSPE). Gleevec induced elevations in P53 and depletion of Bcl2 levels in liver tissues were compared with the control group. Liver sections in rats treated with Gleevec exhibited marked cellular infiltrations, vacuolar degeneration hepatocytes, numerous apoptotic cells, and congestion in central and portal veins, as well as a significant increase in the proliferating of Ki67 after Gleevec injection as compared with control group. In contrast, treatment with Gleevec and GSPE showed a moderate to good degree of improvement in hepatocytes with a significant increase in Ki67, a decrease in P53 and an increase in Bcl2 levels in liver tissues compared to treatment with Gleevec. Therefore, Gleevec induces apoptosis, injury and Ki67 changes in rat liver, whereas GSPE modulates these alternations.

Several functional and morphological studies have been conducted on the pineal gland in many mammalian species; however, no published reports are available on the role of pineal gland on the gonadal development before and after eyelids separation in puppies. Therefore, this study aimed to trace the postnatal histo-morphological changes in the pineal gland and gonads of puppies before (2, 10 and 11 days old) and after (25, 35 and 40 days old) eyelids separation in an attempt to investigate the possible role of pineal gland on the gonadal development. In general, the differentiation of pineal cells, interstitial endocrine cells of testes and stromal ovarian cells coincides with the start of eyelids separation in puppies. Histological examination of stained pineal and gonadal slices of puppies after eyelids separation revealed a remarkable differentiation of pinealocytes and testicular interstitial endocrine cells, as well as presence of some evidence of folliculogenesis in ovary. Surprisingly, melatonin receptor (MT1) protein expression levels were significantly increased in the ovaries and testes of puppies after eyelids separation. Moreover, the mRNA and protein expression of AANAT, a rate-limiting enzyme in melatonin biosynthesis, was notably increased in the pineal gland of opened eyes puppies. Our results suggest an increase of melatonin production from the pineal gland of opened eyes puppies and this could play a vital role in the developmental changes observed in the gonads of these puppies.


Diversos estudios morfológicos y funcionales han sido realizados sobre la glándula pineal en distintas especies de mamíferos. Sin embargo, no hay informes publicados acerca del rol de la glándula pineal en el desarrollo gonadal antes y después de la separación de los párpados en cachorros. Este estudio tuvo como objetivo trazar los cambios histo-morfológicos postnatales en la glándula pineal y las gónadas de los cachorros antes (2, 10 y 11 días de edad) y después (25, 35 y 40 días de edad) de la separación de los párpados, en un intento por investigar el posible rol de la glándula pineal en el desarrollo gonadal. En general, la diferenciación de los pinealocitos, células intersticiales endocrinas de los testículos y las células estromales del ovario coincide con el inicio de la separación de los párpados en cachorros. El examen histológico de glándula pineal y los cortes gonadales de los cachorros, después de la separación de los párpados, reveló una notable diferenciación de los pinealocitos y las células intersticiales endocrinas testiculares, así como la posible evidencia de foliculogénesis en el ovario. Sorprendentemente, en el receptor de melatonina (MT1) los niveles de expresión de proteínas fueron significativamente superiores en los ovarios y los testículos de los cachorros después de la separación de los párpados. Además, el ARNm y la expresión de la proteína AANAT, una enzima limitante de la velocidad en la biosíntesis de la melatonina, aumentaron notablemente en la glándula pineal de los cachorros con los ojos abiertos. Nuestros resultados sugieren que existe un aumento de la producción de melatonina por parte de la glándula pineal en los cachorros con los ojos abiertos, lo que podría jugar un rol vital en los cambios evolutivos observados enlas gónadas de estos cachorros.
